TB-500 research background
Thymosin β4 was isolated from calf thymus in 1981 by Low, Hu and Goldstein and was later found to be one of the most abundant intracellular proteins in mammalian cells, where its principal known function is to sequester monomeric (G-) actin and so regulate cytoskeletal assembly. The actin-binding core is the LKKTET motif at residues 17–23. “TB-500” is a market name, not a chemical one; it is applied both to full-length Tβ4 and to the short synthetic fragment Ac-LKKTETQ, which is a different molecule with a mass near 889 g/mol. The observed mass on the lot COA is the definitive way to tell the two apart.
The published literature on Tβ4 is broad and comes from many independent groups: actin biochemistry, cell-migration assays, corneal and dermal wound models in animals, cardiac models in mice, and a number of formal clinical studies of the full-length protein sponsored by RegeneRx for eye and skin indications, none of which has led to an approved product. These are laboratory findings in cells and animals; they have not been replicated in controlled human trials, and no conclusions about human effects should be drawn from them.
Analytically, full-length Tβ4 behaves more like a small protein than a short peptide. It carries multiple charges and produces a distribution of charge states on electrospray mass spectrometry that must be deconvoluted to the intact mass; it is more prone than short peptides to adsorption on plastic and to freeze–thaw loss; and the single methionine at position 6 is oxidation-sensitive, so a +16 Da satellite peak on the spectrum is the first thing to look for when assessing a lot.
Handling and storage: Lyophilized powder in a sealed vial. Store at −20 °C for long-term storage or at 2–8 °C for short-term storage, protected from light and moisture. Allow the vial to reach room temperature before opening to prevent condensation.
